Probably the steam engine. Newcomen and then later Watt. Really hyper-accelerated human technological advancement (industrial revolution), and thus went on to lift the highest number of people out of poverty in the quickest amount of time to date. It's hard for us moderners to grasp just how exceptionally poor everyone used to be.

Florence was a statistician and a manager. She was officially called a nurse when she became a superintendent of a hospital but had little training in 1853.Less than a year later she was in the Crimea.

From the BBC website,

Florence’s best efforts to combat the rising death toll failed. It kept increasing relentlessly, with over four thousand deaths in a single winter.

Although she had made the hospital more efficient, it was no less deadly. In the spring of 1855, the British government sent out a Sanitary Commission to investigate the conditions at Scutari. It discovered the Barrack Hospital was built on a sewer, meaning patients were drinking contaminated water. The hospital, along with other British army hospitals, was flushed out and ventilation improved. Consequently, the death rate began to fall.

Depends. Sort of chicken or the egg kind of deal.

Is the best invention the "best" or the thing that allowed it to be possible?

Also are we limiting things to direct inventions, or including modifications and discoveries?

Creation of fire was discovered

The first wheels created were not for transportation. They were potter's wheels, then repurposed/modified 300 or so years later for use in wheelbarrows and chariots.

It is a myth. She and a partner did work on patents to block radio signals but these were never employed by the Navy. The technology that led to mobile phones, etc was developed by multiple sources with no use of her patents which were completely unknown, as they'd never been used. A two minute google search will show you the facts.
