For me, so long as the message isn't too full on sex talk wise (that's a no and tells me you lead with your knob) it doesn't matter what you put in a message if I don't like your profile I'm not going to be interested anyway.

My Fab female friends can easily get 10 messages an hour when online. A decent title in the message related to the profile helps, but that just gets me to about 1 in 5 read and 1 in 25 replied to, usually with a ‘no thanks’. Very occasionally it will develop into chat.
