Depends what you're after. If it's good beer and a chat with friends, then I'd say stick to the northern quarter or ancoats.

You’re straight stay away from the Village, too many straight people ruining the place.

The Northern Quarter has some great bars and restaurants, Home Cinema for decent films, Comedy Store for ….Comedy.

If you go Canal Street Manchester, it's easy to get attracted to the big hitters like Onbar, but don't miss out on exploring the sumptuously gothic Via bar
